Abstract:

To provide a method having high performance for the detection of nucleotide polymorphism in a complicate mixture of nucleic acids containing one or more different polymorphs in the mixture and enabling the screening of plural polymorphs at the same time.

The method for judging the identity of polymorphic nucleotide comprises the contact of a target nucleic acid containing a polymorphic site with a solid substrate bonded with one or more gene locus specific primer pairs under a hybridizing condition, the amplification of the target nucleic acid using the gene locus specific primer pairs to obtain an amplification product bonded to the solid substrate at the terminals, the contact of the amplification product with a labeled probe containing at least one detection nucleotide forming a specific base pair with a polymorphic nucleotide in the presence of an enzyme catalyzing the formation of a covalent bond between the detection nucleotide and the amplification product and the detection of the label. The identity of the label on the detection nucleotide shows the complementarity of the polymorphic nucleotide.
