Abstract:

PROBLEM TO BE SOLVED: To obtain the subject new DNA, comprising a base sequence capable of encoding a polypeptide of a glycosaminoglycan sulfotransferase and capable of mass-producing the enzyme useful for related research or the like of the structure and function of the glycosaminoglycan.
SOLUTION: This new DNA has a base sequence, coding a polypeptide of a glycosaminoglycan sulfotansferase, having an amino acid sequence represented by the formula and actions of selectively transferring a sulfate group from a sulfate group donor to the hydroxyl group at the 6-position in an N-sulfated glucosamine residue contained in a glycosaminoglycan which is a sulfate group acceptor, acting on a heparan sulfate or a completely desulfated, N-sulfated (CDSNS)-heparin without acting on chondroitin and chondroitin 4-sulfate, having 6-7 optimal reactional pH and 0.1-0.3 M optimal ionic strength when using NaCl, inhibited with dithiothreitol and adenosine-3',5'-diphosphate and activated with a protamine and is obtained by screening a cDNA library derived from a human fetal brain.
