PURPOSE: To obtain a novel polypeptide used for diagnostic typing of HLA-DR and so on which is related with a DNA sequence coding for the main polymorphic region of the DR β-chain of human lymphocyte antigen(HLA) complex and exhibits immunological and biological activities of the DR β-chain antigen.
CONSTITUTION: Raji cell, of a β-lymphoblastoid cell line is cultured and subsequently the cells are collected. All the RNAs are separated by a conventional method and mRNA is isolated using an oligo(dT) cellulose column. Using this mRNA, cDNA is synthesized and a clone containing a DNA insert related with the HLA-DR-β-chain is selected. This is linked to an expression control sequence to obtain a recombinant DNA molecule containing DNA insert comprising the DNA insert DR-β-C, the DNA hybridizing the DNA insert and the DNA having nucleotide sequence coding on the expression for the polypeptides coded for either of the DNA above or the like. The recombinant DNA molecule is transduced into a host to transform it and the transformant is cultured to obtain the polypeptides related with the DNA sequence coding for the HLA-DR-β-chain.
