Abstract:
PURPOSE: To obtain an immunoreactive composition comprising an immunoglobulin covalently bonded to a low-molecular weight polysaccharide or its antigen recognizing fragment and having reduced immunogen performance or improved medicine action. CONSTITUTION: A hydroxyl group of a polysaccharide having low-molecular weight, preferably a reactive hydroxyl group of dextran having 3,000-16,000 molecular weight, especially 6,000-16,000 molecular weight is oxidized into an active aldehyde group in the presence of an oxidizing agent, preferably sodium periodate to activate the polysaccharide and mixed with desired immunoglobulin in a proper solution (e.g. buffer solution having pH6.5-7.5) and aldehyde group of oxidized dextran is reacted with amino group of the immunoglobulin to form a Schiff base adduct. The Schiff base adduct is reduced with a proper reducing agent (cyanoboron hydride sodium) to provide the objective dextran- immunoglobulin adduct of substantially monomer.
