Abstract:
PURPOSE: To obtain an aromatic ether useful as an intermediate for an adrenergic β-blocker at a high yield without producing any by-product by allowing propanediol sulfate to react with phenol.
CONSTITUTION: A 3-halo-1,2-propanediol sulfate of formula I (wherein X is Cl, Br) is allowed to react with a phenol of the formula; Ar-OH (wherein Ar is an aromatic hydrocarbon, an aromatic heterocyclic ring) in the presence of a base (e.g. NaOH) in a solvent such as acetonitrile at 0 to 80°C to obtain a compound of formula II and formula III (wherein M is H, an alkali metal, ammonium ion) which does not need to be isolated as an intermediate. Then, this compound is filtered and separated to obtain the target compound of formula IV.
