PURPOSE: To obtain the titled factor containing no erythropoietic inibitory factor in large amounts, by adding ethanol to a solution containing erythropoietin in two stages, removing a precipitate formed in the first stage, and recovering the resultant precipitate formed in the second stage.
CONSTITUTION: A solution containing erythropoietin, e.g. urine, blood plasma or placental extract, is adjusted to 5W15mg/ml with preferably an aqueous solution of 1W5M LiCl and adjusted to 5W8pH, and ethanol is added thereto under cooling with ice to give 65W75% final concentration. The formed precipitate is then removed by centrifugation, and ethanol is then added to the residual supernatant liquid under cooling with ice to give 85W95% final concentration. The resultant precipitate is then recovered by centrifugation. The recovered precipitate is further dissolved in a small amount of distilled water and subjected to the gel filtration with "Sephadex" t isolate the aimed erythropoietin of good quality. The erythropoietin is an essential factor for differentiation of blood stem cells which are the cell regeneration systems into erythrocytic cells.
