PURPOSE: To simply obtain a heat resistant restriction enzyme having high active purity in high yield by subjecting a restriction enzyme solution to heat treatment for a time when an endonuclease contained as impurity is substantially deactivated at the lowest temperature at which an objective heat resistant restriction enzyme starts to deactivate.
CONSTITUTION: A restriction enzyme-containing solution is subjected to heat treatment at the lowest temperature at which the objective heat resistant enzyme starts to deactivate for a time when an endonuclease contained as an impurity is substantially deactivated. The solution is subjected to heat treatment at 60°C for 10min in the case of a new heat resistant restriction enzyme Sel I (whose enzymatic properties are shown in the table) derived from thermophilic Synechococcus elongatus (FERM P12393).
