Abstract:

PURPOSE: To obtain the titled compound useful as a clinical diagnostic, etc., economically in an industrial scale, by removing impurities from an aqueous solution of amorphous NAD with a weakly basic ion exchange resin, adjusting the concentration of the purified solution, adding a crystallization nucleus thereto, cooling the solution to effect the crystallization, separating the crystal, dissolving in water, and freeze-drying the solution.

CONSTITUTION: An aqueous solution of amorphous β-nicotinamide-adenine-dinucleotide (NAD) is treated with (A) an acetate-type highly porous weakly basic anion exchange resin to remove the impurities. (B) The concentration of the purified solution is adjusted to 20W60%. (C) A crystal is added to the solution as a crystallization nucleus. (D) The solution is cooled to 0W20°C. The crystal of free acid-type β-nicotinamide-adenine-dinucleotide tetrahydrate is crystallized by the combination of the steps (A)(B)(C)(D), (A)(B)(D) or (B)(C)(D), and the crystal is dissolved in water. The solution is subjected to the freeze-drying, or is precipitated with an alcohol and dried to obtain the objective compound.
