To provide a method for preparing leukocytic α-interferon, intended for raising the productivity of α-interferon per donor by increasing leukocyte levels as well as the production of specific α-interferon.
This method for preparing leukocytic α-interferon comprises separating leukocytes from human blood, purifying the leukocytes through hemolysis with ammonium chloride followed by washing with a physiological saline buffer solution, suspending the purified leukocytes in a liquid medium, inducing α-interferon by Sendai virus followed by performing a pretreatment at 30-40°C for 60-90 min, continuing a culture at ≤30°C, adding additional Sendai virus to the suspension at the end of the culture, and then separating the cells followed by adjusting the resultant supernatant to >pH 7.
