Abstract:
PURPOSE: To realize the preparation of microgranules smaller than a specific grain size and having rich suspensibility to liquid by using a high shear mixer- granulator to mix and knead active ingredients and an excipient by spraying bonding liquid when preparing microgranule such as sustained release liquid medicinal preparations or the like.
CONSTITUTION: When preparing spherical microgranules with sizes smaller than 500 μm and surfaces suitable for coating, a high shear mixture-granulator provided with a mixer and a mill is used to mix active ingredients and suitable excipient in the granulator. Further, at this time, the mixture is wetted by providing 80 to 180 g of bonding liquid (granulating liquid) per kilogram of the powder mixture based on predetermined parameters. Specifically, the bonding liquid is sprayed to the mixture at a spray rate of 10 to 40 g/min and a spray pressure of 1.5 to 2.5 bar. Then, such a mixture provided with bonding liquid is kneaded at a mixer rotational speed of 50 to 600 rpm and a mill rotational speed of 500 to 400 rpm, thus obtaining the required microgranules.
