Abstract:
PURPOSE: To prepare a polypeptide in vertebrate host cell culture system by transforming a vertebrate cell culture with a replicable expression vehicle in a specific form having DNA sequence encoding a polypeptide.
CONSTITUTION: (a) DNA Transfer vector fragment replicable in a host cell culture is combined with (b) a DNA fragment containing a promoter which is adapted to the host cell culture and (c) a DNA encoding polypeptide, suitable for binding to the fragment (b), but heterogenous to the fragment (b) together with a transfer termination site arranged at a proper position, poly A addition sequence, initiation signal of translation and termination signal to form a replicable expression vector. The vertebrate cell culture is transformed by the vector. The C Sequence of the expression vector is under control of a promoter and a translation initiation codon is arranged within ±1 base pair of original position at which translation initiation codon of protein is located.
