Abstract:

PURPOSE: To prepare the titled substance useful as an adrenocortical hormone preparation, etc. economically, in an industrial scale, by treating the 17α,21-cyclic orthoester of a specific 17α,21-dihydroxy-20-keto-pregnane steroid with an ion exchange resin.

CONSTITUTION: The 17α,21-cyclic orthoester of 17α,21-dihydroxy-20-keto-pregnane steroid of formula I (R is hydrocarbon group; R' is lower alkyl), e.g. hydrocortisone 17,21-ethyl orthopropionate [ 17α,21-( 1'-ethoxy-1'-ethyl-methylenedioxy )-11β-hydroxy-4-pregnene-3,20-dione] etc. is dissolved in methanol, etc., and diluted with water purified with an ion exchange resin. A cation exchange resin is added to the solution and stirred. After reaction, the resin is separated, and the solution is distilled under reduced pressure to distil out the solvent and to obtain the objective substance of formula II.
