PURPOSE: To prepare the titled compound useful as a remedy for cerebral thrombosis, economically, in high yield, by reacting a reaction product of theobromine and 1-bromo-3-chloropropane successively with an alkali iodide and acetylacetone.
CONSTITUTION: 1-(3-Chloropropyl)theobromine of formula I is prepared by reacting theobromine with 1-bromo-3-chloropropane in the presence of an equimolar amount or slightly excessive alkali carbonate or alkali hydroxide in a water-soluble organic solvent or a mixture of the solvent and water. The compound of formula I is made to react with an alkali iodide in an organic solvent to afford 1-(3-iodopropyl)-theobromine of formula II, which is converted to the objective compound of formula III by reacting with acetylacetone in the presence of an alkali carbonate or an alkali hydroxide in a lower alkanol.
