Abstract:
PROBLEM TO BE SOLVED: To selectively obtain the subject compound useful as e.g. a substrate for assaying α-amylase activity, by reaction of lactose with 2-chloro-4- nitrophenyl-α-maltoside in an aqueous medium in the presence of β-galactosidase.
SOLUTION: This compound, 2-chloro-4-nitrophenyl 4-O-β-D- galactopyranosyl-α-maltoside of formula II useful as e.g. a substrate for assaying α-amylase activity, is obtained selectively by reaction of lactose with 2-chloro-4- nitrophenyl-α-maltoside of formula I in water or a solution as a mixture of water and a hydrophilic organic solvent (e.g. isopropyl alcohol) in the presence of β-galactosidase derived from Bacillus circulans at a concentration of 0.01-0.8mg per mL of the above solution.
