Abstract:

PURPOSE: To readily obtain the title optically active substance by hydroxylation, using microorganisms such as of Streptomyces sp., of 4-(4-cyanoanilino)-6,7- dihydro-5H-cyclopenta[d]pyrimidine.

CONSTITUTION: Microorganisms classified as Streptomyces or Cunninghamella sp., is put to culture under aerobic conditions in a medium containing nutrients such as carbon and nitrogen sources. And at a point elapsed 1-5 days after the beginning of the culture, a raw material compound is added to the medium and put to culture for ca. 1-14 days. Alternatively, the bacteria is put to culture and collected, the resultant bacteria being brought into contact with the raw material compound. Thence, after completion of the conversion reaction, the resulting product is filtered, the resultant filtrate being extracted with an organic solvent poorly miscible with water such as ethyl acetate followed by separation to carry out purification, thus obtaining the objective (7S)-4-(4-cyanoanilino)-6,7- dihydro-7-hydroxy-5H-cyclopenta[d]pyrimidine of the formula.
