PURPOSE: To produce the subject active ingredient at reduced manufacturing cost industrially and advantageously by culturing callus of tissue culture of TOCHU (Eucommia ulmoides) first by a callus multiplication process at a specific illuminance and by an active ingredient production process at a specific saccharide concentration of medium.
CONSTITUTION: Part of root, leaf or stem of TOCHU (Eucommia ulmoides) is cut off, inoculated into an agar medium, cultured for about 30 days to form callus of tissue culture of TOCHU (Eucommia ulmoides), which is subjected to liquid culture in a flask to prepare a seed callus for culture in a large amount. The seed callus is transplanted in a large-sized tank culture device in a callus culture process in a large amount, the callus is cultured in a large amount at ≥3,000lux illuminance in the first callus multiplication process and, in the second active ingredient production process, a saccharide is added to the callus cultured in a large amount and the callus is cultured at ≥2% saccharide concentration of medium to produce the active ingredient, the active ingredient is extracted from the callus containing a large amount of the active ingredient in a component extracting and separating process and passed through a forming and separating column to give the active ingredent such as pinoresinol diglucoside, etc.
