Abstract:

PURPOSE: To obtain an alkaline protease useful for leather processing, food industry, detergent, etc., in high efficiency by culturing a microbial strain transformed with an expression vector containing an alkaline protease gene originated from a microorganism of the genus Bacillus and having a specific restriction map.
CONSTITUTION: This alkaline protease useful for leather processing, food industry, detergent additive, pharmaceuticals, etc., is produced by separating a chromosomal DNA from a microorganism of the genus Bacillus (e.g. Bacillus licheniformis ATCC-14580), subjecting the DNA to restriction enzyme treatment, integrating into a cloning vector, cloning a microorganism to obtain a transformed microorganism containing a structure gene coding alkaline protease and transformed by the introduction of a DNA fragment containing a region including an alkaline protease structure gene restricted both terminals with restriction enzyme cleavage points HpaI and HaeIII (2), containing other restriction enzyme cleavage points Hind and SphI between the regions and containing a DNA fragment having a restriction map expressed by the formula, culturing the obtained microorganism and separating the enzyme from the cultured product.
