PURPOSE: The reaction between a monobromochlorophenol and ammonia in the presence of a copper catalyst is conducted in a specific temperature range to produce the titled compound that is used as a medicine by usual purification operations in high yield.
CONSTITUTION: A monobromochlorophenol such as 2-chloro-4-bromophenol is mixed with ammonia and they are made to react in the presence of a copper catalyst such as powdered copper with heat at 50W120°C to produce the titled compound. As an ammonia source, is used its aqueous solution of higher than 28% concentration or liquid ammonia and the amount employed for the reaction is usually more than 5mol/mol of the monobromochlorophenol. The reaction is effected in a tightly sealed reactor under the autogeneous pressure caused by heating.
EFFECT: Bromine atoms are selectively substituted with amino groups. The reaction is mild.
