Abstract:

PURPOSE: To obtain the subject compound useful as an intermediate for synthesizing pharmaceutical compounds such as guanine nucleotide analogues by reacting guanine with a chlorinating agent in the presence of a dialkylaniline.
CONSTITUTION: Guanine is reacted with a chlorinating agent (e.g. phosphorus oxychloride) in the presence of a dialkylaniline (e.g. N,N-diethylaniline) preferably in a solvent-free state at 70-120°C for 20min to several hr to obtain 2- amino-6-chloropurine of the formula. The chlorinating agent and the dialkylaniline are used in amounts of 3-10 times moles and ≥2 times moles, respectively, the amount of the guanine.
