Abstract:

PURPOSE: To obtain the subject enzyme useful for measuring benzylamine, etc., which is an index of decomposition metabolism of in vivo amines by culturing a microorganism, belonging to the genus Enterobacter and capable of producing a benzylamine transaminase and collecting the microorganism having the ability to produce the benzylamine transaminase from the prepared culture.
CONSTITUTION: A microorganism, belonging to the genus Enterobacter, Bacillus, Agrobacterium, Corynebacterium, Comamonas, Pseudomonas, Brevibacterium, Streptomyces, Actinomyces, Spirillospora, Penicillium or Neocosmospora and having the ability to produce a benzylamine transaminase [e.g. Bacillus brevis B-175 (FERM P-13312)] is inoculated into a culture medium and cultured at 30°C for 36hr under shaking and the resultant culture solution is subsequently centrifuged to collect a microbial cell pellet, which is then suspended in a 0.1M phosphoric acid buffer solution. Ultrasonic waves are subsequently applied thereto to crush the microbial cells. The crushed microbial cells are then centrifuged to collect the supernatant, which is subsequently purified by a method for precipitation with ammonium sulfate, ion exchange column chromatography, etc., to efficiently afford the objective enzyme.
