Abstract:
PURPOSE: To obtain a new plasmid vector capable of expressing in Escherichia coli and/or Bacillus subtilis and useful for obtaining a transformed microorganism capable of producing β-lactamase as well as a protein such as hormone or protein for foods in a high yield.
CONSTITUTION: This plasmid vector arranges a gene coding for a heterologous protein under the control of an erythromycin promoter of pE194 modified by replacing the -35 region with a terminating sequence of phage 5d of Escherichia coli and a synthetic DNA segment of a base sequence of formula I or formula II. A microorganism transformed by plasmid vector pSM146 and pSM147 in the case in which the above gene is β-lactamase gene of pBR322 of Escherichia coli is Escherichia coli (pSM146)ATCC 53039 and Escherichia coli (pSM147) ATCC53040 and produces β-lactamase. The preparation of the plasmid vector is carried out according to the well-known method.
