Abstract:
PURPOSE: To provide the method for simply and efficiently producing a foreign DNA gene bank using a cosmid vector produced by a genetic engineering technique.
CONSTITUTION: The method for producing the gene bank of the foreign DNA comprises cleaving the DNA of a lambda phage or lambdoid phage having the same directional property and having two cos units with two kinds of restriction enzymes, inserting various kinds of foreign DNA fragments having lengths of 35-45Kbp between the cleaved right and lest arms having the cos units, respectively, converting the treated DNA fragments into a transduced phage particle by an in vitro packaging method, and subsequently cloning the transduced phage particle in Escherichia coli.
