PURPOSE: To obtain an expression vector capable of expressing a DNA sequence encoding human insulin-like growth factor in a transformed recombinant bacterial host, and capable of producing the subject material useful for treatment or the like for the symptom affecting the growth by a recombinant DNA technique.
CONSTITUTION: This method for efficiently producing a matured protein of a human IGF by a recombinant DNA technique comprises a step for recovering pBR322 plasmid containing a promoter region and derived from Escherichia coli or the like as a vector capable of being expressed in a prokaryotic cell, a step for treating the pBR322 plasmid with a restriction enzyme, a step for incorporating a DNA encoding human insulin-like growth factor(IGF) to prepare a recombinant plasmid containing the human IGF gene, a step for inserting the recombinant plasmid into a transformed recombinant prokaryotic host such as E. coli-K12 strain 294 (ATCC No.31446) to transform the host, and a step for culturing the transformant by the expression vector capable of expressing a DNA sequence encoding the human IGF in the prokaryotic host.
