PURPOSE: To obtain a recombinant HIV-1 antigen and to use it as a diagnostic by transducing Escherichia coli with Escherichia coli β-galactosidase of HIV-1 specific protein part or a vector plasmid fused with the part.
CONSTITUTION: The projected end of Asp718 cDNA fragment of 25,716p from SalI-SstI cDNA fragment (PHIVenv) of 3,750bp of HTLVIII separated strain BH10 is filled with PolI, cleft by HindIII and filled with PolI to give pBD2, which is cloned to give pMB2440. The plasmid codes a part of HIV envelop protein having a seqeunce of a joint part between gp120 and gp41 and is suitable for preparing an antigen. pHIV core of BH10 is digested with PvuII and XbaI to give a cDNA fragment of about 1,350bp, which is digested with SmaI and XbaI to give pBD21C20H, which is cloned to give a pCO1. The plasmid is used to produce a fusion protein for detecting HIV-1 antibody in serum.
