Abstract:

To obtain the subject substrate improved in stability of an enzyme substrate for enzyme activity measurement and enzymeimmunoassay when preserved for a long period of time, by bringing a solution of an enzyme substrate such as a hydrolase into contact with a carrier capable of scavenging a metal ion to remove the metal ion.

2-Amino-2 methyl-1-propanol and sodium azide are weighed in a container, are mixed with purified water, incorporated with concentrated hydrochloric acid by monitoring by a pH meter, adjusted to pH10, diluted with purified water and mixed with 4methylumbelliferone phosphate (4-MUP) to give a 1mM 4-MVP solution (black dots). Then an iron solution is added to the MVP solution so as to give 10ppm concentration. The prepared solution is used as a reference solution (black triangles), is passed through a chelate resin column and subjected to a metal ion removal treatment to give a column treated substrate solution (black square). The solution is examined by a preservation stability test by an incubator at 4°C. Consequently, the acquisition of the objective enzyme substrate preventing deterioration in quality is observed.
