Abstract:
PROBLEM TO BE SOLVED: To obtain an intramolecularly coupled compound by reaction of a phenol derivative with a hypervalent iodine (III) reagent, and to obtain the subject alkaloid by using the above compound.
SOLUTION: This intramolecularly coupled compound of formula II is obtained by reaction of a phenol derivative of formula I (R1 and R2 are each H or a hydrocarbon; R3 is an organosilicon group; R4 is an organic oxygen- contg. group) with a hypervalent iodine(III) reagent, phenyl iodine(III)bis(trifluoroacetate) in a solvent such as trifluoroethanol at -50 to 10°C. A galanthamine-type alkaloid of formula III is obtained, for example, by deprotection of the catechol site of the above intramolecularly coupled compound followed by methylating the phenolic hydroxyl group. By using the hypervalent iodine reagent, the other objective galanthamine-type Amaryllidaceae alkaloid derivative can be easily obtained.
