Abstract:
PURPOSE: To produce a laxative composition in a unit dosage form capable of achieving a high dose in a pharmaceutical preparation of a small physical size by melting a stool softener, then mixing a stimulant laxative compound therewith and filling the resultant mixture in a capsule.
CONSTITUTION: A normally solid stool softener (e.g. poloxamer) is melted and a stimulant laxative compound (e.g. danthron) is then dispersed and/or dissolved in the molten stool softener and mixed therewith to prepare a homogeneous mixture. The resultant molten dispersion is then filled in a hard gelatin capsule shell and the capsule is further cooled to solidify the melt or dispersion and afford the objective substance. The poloxamer and the danthron are used at a weight ratio of the poloxamer to the danthron of (2:1) to (20:1), most preferably (8:1) to (13.3:1). Furthermore, the volume of the active ingredient can be minimized without adding an excipient or another adjuvant thereto at all. An unexpectedly high dose can be achieved with a pharmaceutical preparation of a small physical size.
