Abstract:

PROBLEM TO BE SOLVED: To simply obtain the subject block useful for preparing a DNA oligomer without passing through a synthesis process from a nucleotide and isolation and purification processes by oxidizing or sulfurizing a trivalent phosphorus atom of the nucleotide prepared by a specific method into a pentavalent phosphorus atom.
SOLUTION: A nucleotide derivative of formula I (B1 and B2 are each a base to be protected with a protecting group ordinary in nucleotide chemistry; R1, R2, R1' and R2' are each H or the like; R3 is a protecting group; A1 and A2 are each OH or the like; X is azolyl; Z is O or S; n is 0 or 1; with the proviso that a case in which combinations of R1 and R2 and R1' and R2' are H and H is omitted) is reacted with a 3'-O,5'-O-nonprotective nucleotide derivative of formula II (B3 is as shown for B1 or the like; A3 is as shown for A1 or the like; Z' is as shown for Z) or a nucleotide derivative to give a nucleotide derivative of formula III. The trivalent phosphorus atom of the nucleotide is oxidized or sulfurized into a pentavalent phosphorus atom to give the objective block of formula IV.
