Abstract:

PROBLEM TO BE SOLVED: To efficiently obtain an optically active amine derivative having biological actions such as affinity to a melatonin receptor and useful as a medicinal agent or a raw material for synthesizing medicinal agents by an assymmetric reduction method using rutenium-optically active phosphine complex as a catalyst.
SOLUTION: A wanted optically active form of a compound expressed by formula II is obtained by reacting an (E) form expressed by formula I {R1 and R2 are each H, a (substituted) hydrocarbon, etc.; R3 is H or an acyl; X is (CH2)n, [(n) is 1-4], N, O, S, (m) is 1-5: A is a (substituted) ring or forms a condensed ring with a five or six membered ring; R3 is acyl when (m) is 1} and a (S)-form which is an optical isomer of the same with a compound of formula I having 1-500mg/ml concentration using either one of optical isomers having (R) or (S) configuration of optically active tert. phosphine in a ruthenium- optically active phosphine complex as a catalyst in about 0.1 millimol-1mol catalyst concentration based on 1mol of the compound of formula I, at 15-100°C and 5-100atm steam pressure for about 1-48 hours.
