Abstract:

PROBLEM TO BE SOLVED: To obtain the subject derivative useful as a high degree sweetener, etc., by subjecting a sucrose alkyl 4,6-orthoacylate to mild aqueous acidic hydrolysis to provide a mixture of 4- and 6-monoesters of sucrose and then treating the ester mixture with a base to convert the sucrose 4-ester into sucrose 6-ester.
SOLUTION: The sucrose 6-acylate of formula II useful as a key intermediate in the preparation of sucralose is prepared by subjecting a sucrose alkyl 4,6- orthoacylate of formula I (R4 is an alkyl; R2 is an alkyl or an aryl) (for example, sucrose methyl 4,6-orthoacylate) to mild aqueous acidic hydrolysis in the presence of an acid catalyst (for example, p-toluene sulfonic acid) in an inert polar organic solvent (for example, dimethylformamide) containing water in a larger amount than a theoretically required amount to provide a mixture of sucrose 4-and 6-acylates and then treating the mixture with a base such as a tertiary amine (for example, pyridine) to convert the 4-acylate to the 6- acylate.
