Abstract:
PURPOSE: To efficiently separate a produced inorganic salt and a urethane compound due to the reaction of an inorganic base by reacting an amino acid ester hydrochloride with a dicarbonate in the presence of the inorganic base in a specified organic solvent.
CONSTITUTION: An amino acid ester hydrochloride of formula I [X is amino acid residue; R2 is alkyl, alkenyl or aralkyl; (n) is ≥1] is made to react with a dicarbonate of formula II (R1 is alkyl, alkenyl or aralkyl) in the presence of an inorganic base [e.g. NaOH or Ca(OH)2] in an organic solvent to provide a urethane compound of formula III (Am is residue after removing amino group from an amino acid ester). In the process, an organic solvent (e.g. any of isopropyl alcohol, tetrahydrofuran, dioxane, acetonitrile, toluene and dichloromethane or a mixture thereof) hardly dissolving the inorganic salt formed by neutralizing the inorganic base during the reaction and capable of dissolving the produced urethane compound is used as the organic solvent. The salt formed as a by-product can readily be removed by filtration, etc.
