PROBLEM TO BE SOLVED: To provide a purification method for separating a polypeptide of interest (e.g., an antibody) from one or more contaminants.SOLUTION: A method for purifying a polypeptide by ion exchange chromatography comprises changing the conductivity and/or pH of buffers; specifically, binding the polypeptide of interest to an ion exchange material at a first conductivity and pH, and then washing the ion exchange material with an intermediate buffer having a different conductivity and/or pH.SELECTED DRAWING: Figure 3
