Abstract:

PURPOSE: To purify an enzyme as penicillin amidase with a high combining ability with industrial advantage by specifically immobilizing an enzyme opposing to the substrate bearing an aromatic group in aqueous solution to a specific carrier substance to which phenylbutylamine groups are covalently bonded.

CONSTITUTION: An enzyme as penicillin amidase that opposes to the substrate bearing aromatic group and is in an aqueous medium is brought into contact with a carrier substance T-pa that is prepared by allowing phenylbutylamine to react with the glycidyl group of the carrier precursor T-Vs-A made of crosslinked copolymer comprising acrylamide, glycidyl methacrylate and N,N'- methylene-bis-methacrylamide thereby specifically immobilizing the enzyme through hydrophobic bonds to purify the enzyme.
