Abstract:

PURPOSE: To recover kallikrein from human urine, in high yield and purity, economically, by using a water-insoluble carrier containing arginine, agmatine, lysine or benzamidine, etc. as a ligand.

CONSTITUTION: Fresh undecomposed human urine is made to contact with colloidal hydrated aluminum silicate adsorbent at 4.0W6.0pH and room temperature, preferably at 5W30°C to effect the selective and complete adsorption of proteases other than kallikrein. The supernatant liquid is separated by decantation or centrifugal separation, adjusted to 6.5W9.5pH, preferably 8W9.5pH, and brought into contact at room temperature, preferably 5W30°C with a carrier having an affinity with kallikrein and obtained by bonding arginine, agmatine, lysine or benzamidine, etc. to a water-insoluble carrier such as agarose to effect the specific adsorption of kallikrein. The adsorbed kallikrein is eluted with eluting buffer solution.
