To inexpensively obtain the subject α-amylase useful for conversion, etc., of starch to saccharides and stable to acids by heating an enzyme- containing solution derived from fungus to inactivate glucoamylase and removing the modified enzyme.
In an enzyme-containing solution comprising fermentation process, etc., of fungus of the genus Aspergillus, preferably Aspergillus niger or Aspergillus awamori, the pH of the solution is controlled to 1-8 and the solution is heated at 40-80°C, preferably 50-75°C which is a temperature enough to inactivate glucoamylase for a time enough to inactivate the glucoamylase in the presence of calcium ion and the modified glucoamylase is removed to provide the objective α amylase useful for converting starch to high-maltose syrup or highmaltotriose syrup, etc., containing glucose and stable to acid.
