Abstract:

PROBLEM TO BE SOLVED: To provide a method and a reagent for detecting a trace amount of protein, capable of easily and quickly detecting and/or measuring basic protein of an urophanic disease marker, such as the Bence-Jones protein(BJP) useful as a diagnostic marker for multiple myeloma with high sensitivity.
SOLUTION: This method and its measurement reagent set includes a process for extracting and separating the BJP inside a urine sample from other proteins in urine by aqueous two phase partition using water soluble polymer and cation modified polymer, a process cleaning an extracted phase, containing the BJP of an objective protein, and a process reacting the extracted BJP with a pigment. Coloration of a BJP-pigment combination can be qualitatively and/or quantitatively analyzed and evaluated by a sensitizing function under coexistence of the water soluble polymer. The basic protein, belonging to prerenal protein with a high isoelectric point such as the disease diagnostic marker BJP, can be detected and measured easily and quickly with high sensitivity by solid phase extraction/pigment bonding.
