Abstract:
PURPOSE: To obtain the subject new compound having potent cyclic GMP-specific phosphodiesterase-inhibitory action, and useful for treating hypertension, heart failure, myocardial infarction, arteriosclerosis, asthma, atopic dermatitis, etc.
CONSTITUTION: This compound (salt) is expressed by formula I (R1 is H, methyl, etc.; R2 is a 1-4C alkyl; (n) us 0 or 1; X is a halogen, phenoxy, etc.), e.g. 2-[5-(chloroacetamido)-2-ethoxyphenyl]quinazolin-4(3H)-one. This compound of formula I is obtained, for example, by the following process: a 2- aminobenzamide of formula II is reacted with a compound of formula III in the presence of an organic base such as triethylamine followed by treating the reaction product with an inorganic base such as NaOH and then reducing the nitro group of the product with a reducing agent such as nickel chloride- sodium borohydride to form a compound of formula IV, which is then reacted with a compound of formula, ClCO(CH2)nZ (Z is a halogen, phenoxy, etc.) in the presence of an organic base.
