Abstract:
PROBLEM TO BE SOLVED: To provide an RNA amplification method which can stably detect HIV-1 in which genome mutation occurs easily, at a temperature condition of 41°C or less where it is easy to heat.SOLUTION: As a result of the detail analysis of the mutation reported in HIV-1, we found out that in an HIV-1 genome, an RNase H genome region has less mutation than other regions have. Thereby, RNA is amplified using two kinds of DNA primers which can specifically bind to both ends of the amplification gene moiety of a target gene, a reverse transcriptase, and an RNA polymerase, with all or a part of human immunodeficiency virus type I (HIV-1) RNase H DNA as a target.SELECTED DRAWING: Figure 1
