Abstract:
PROBLEM TO BE SOLVED: To provide a method for identifying host genes and encoded proteins for therapeutic targets in a short period of time.SOLUTION: The period for identification of promising targets is reduced from months to weeks or less by the combination of Random Homozygous Gene Perturbation (RHGP), that a Gene Search Vector, that is either lentivirus or MMLV-based, is inserted into a cell genome, with Thermal Asymmetric Interlaced (TAIL) PCR for the vector insertion site. RHGP is used to identify potential host targets for influenza, HIV and other viral infections. Specific targets including PTCH1, Robo1 and Nedd4 are reviewed in detail.
