Abstract:

PURPOSE: To obtain a reactive microsome-forming agent capable of introducing a polyalkylene oxide chain into a microsome such as liposome and further capable of immobilizing a functional substance at the tip of the chain, and to prepare the microsome containing the reactive microsome-forming agent.
CONSTITUTION: This reactive microsome-forming agent comprises a reactive phospholipid derivative of formula I [R1C(=0), R2C(=0) are the acyl residue of a 3-30C fatty acid; R3 is H, methyl; AO is a 2-4C oxyalkylene; (n) is an average oxyalkylene addition molar number of 1-1000; Y is group of formula II (p) is 2, 3), group of formula III ((q) is 0-3); M is H, an alkali metal]. The compound of formula I is obtained e.g. by a method comprising reacting a polyalkylene glycol derivative having a carboxyl group at one end and a hydroxyl group at the other end with a dicyclohexylcarbodiimide to activate the reaction system, reacting the produced active type compound with a phosphatidyl ethanol compound, and subsequently reacting the obtained compound with N, N'-carbonyl di-midazole.
