Abstract:

PURPOSE: To readily measure the titled enzyme useful for screening patients suffering from hepatopathy and hematopoietic malignant tumor in blood serum with a high accuracy, by adding an inhibitor capable of simultaneously inhibiting activities of allylamidase and cystylaminopeptidase.

CONSTITUTION: An inhibitor for allylamidase (AA) and cystylaminopeptidase (CAP), e.g. o-phenanthroline, etc., glutamic acid dehydrogenase, reduced form nicotinamide adenine dinucleotide phosphate (NADPH), α-ketoglutaric acid, magnesium chloride and a buffer solution at 7.0W8.0 pH are added to a blood serum specimen and incubated at 37°C for 5W10min. A buffer solution at 7.0W8.0 pH containing L-leucinamide as a substrate for cytosol leucine aminopeptidase (CLAP) is added to measure the reduction of absorbance of the NADPH in the ultraviolet region. Thereby the cytosol leucine aminopeptidase is specifically measured.
