Abstract:

PURPOSE: To obtain a trypsin inhibitor useful as e.g. an anti-inflammatory agent, by bringing human urine into contact with a styrene-based anion exchange resin at a specific pH level followed by eluting the resultant adsorbate with an aqueous alkaline solution and then treating the elute with an -polylysine-immobilized insoluble carrier to elute the adsorbate.

CONSTITUTION: First, a 3N aquaous NaOH solution is dripped into human urine such as healthy adult man's urine to adjust the urine to pH8.5, the insolubles formed is filtered off, 3N hydrochloric acid is then added to the filtrate under agitation to adjust the filtrate to pH6.0-8.0, the resultant filtrate is brought into contact with a styrene- based anion exchange resin to effect adsorption of a trypsin inhibitor in the urine to the resin followed by washing the adsorbed body with a 0.5% aqueous ammonium borate solution (pH7.0) and treatment with a 0.5M ammonium chloride-aqueous ammonium solution (pH9.5) to effect eluting the adsorbate, and the resultant elute is brought into contact with an -polylysine-immobilized insoluble carrier to make the adsorbate adsorb to the carrier followed by washing the adsorbate with an aqueous sodium phosphate solution and then eluting the absorbate with a 0.1M phosphate buffer solution (pH7.0), thus obtaining the objective compound, a trypsin inhibitor, to be used as e.g. an anti-inflammatory agent in high purity and yield.
