Abstract:
PURPOSE: To prepare a stereospecific retinoid derivative applied for the local and systemic treatment in the condition of skins connected to the disease of hyperkeratosis and/or the conditions of skins having immunogenic allergic reaction and the degenerative diseases of connected tissues.
CONSTITUTION: The stereopsecific retinoid derivative of formula I (R is H or a 1-4C thioalkyl), such as the all-trans retinoid derivatives of the formula II and formula III and the 13-cis-retinoid derivative of formula IV. The compd. is obtd. by bringing dicyanpropenal of formula V to react with ethynyl-β-ionol of formula VI in the presence of an organic magnesium compd., hydrogenating the acetylenic linkage of the obtd. compd. of formula VII (the use of aluminum- lithium hydride is sutltable) to obtain a compd. of formula VIII and then reducing the compd. by using titanium having low valences. The compds. of the formula I to the formula IV are hydrolized, by which the same stereospecific retinal or retinoic acid can be obtd.
