Abstract:

PURPOSE: To obtain a new compound having excellent radiation damage- protecting action.

CONSTITUTION: A compound of the formula (R1 is an amino which may be substituted with a 1-6C alkyl, etc.; R2 is H or a 1-6C alkyl; R3 is a 1-6C alkyl or R2 and R3 together represent a 1-8C alkylidene), e.g. methyl-3-deoxy-3-amino-5- deoxy-5-thio-D-ribofuranoside. The compound is obtained by selectively protecting, e.g. a hydroxyl group of 1,2-O-isopropylidene-α-D-xylose with trithyl chloride, etc., then reacting a hydroxyl group at 3 position with acetic anhydride in the presence of dimethyl sulfoxide, reacting the product with hydroxylamine to give a 3-oxime substance, reducing the 3-oxime substance with lithium aluminum hydride and successively acetylating the produced 3-amino substance, deprotecting the 5-position and further activating the deprotected compound with paratoluenesulfonic acid, etc., reacting the activated compound with potassium thioacetate and finally deacetylating 5 position of the reaction product.
