Abstract:
PURPOSE: To produce sarcosine oxidase at a high productivity without requiring any inducing substance in a medium in the production.
CONSTITUTION: The objective enzyme catalyzing an enzyme reaction exhibits following physicochemical properties and activities of catalase, creatinase and N-ethylglycine oxidase as admixed enzymes of: ≤0.05 unit catalase activity, ≤0.0004 unit creatinase activity and ≤0.03 unit N-ethylglycine oxidase activity. (a) Action: generates glycine, formaldehyde and a molecule of hydrogen peroxide from sarcosine, a molecule of oxygen and a molecule of water, (b) substrate specificity: has a substrate specificity in sarcosine, (c) optimum pH: 8.0-9.5, (d) isoelectric point: 4.7, (e) molecular weight: 4⊗104 (measured by gel filtration technique) and (f) heat stability: stable to 10min treatment.
