Abstract:

PURPOSE: To obtain a novel vector used as the label for selecting only a transformer holding a recombinant into which an extraneous DNA fragment is integrated from among colonies of the transformer.

CONSTITUTION: A vector, e.g. plasmid pERISH1/JM109 (FERM P-11644), having a region of rcsB gene which is a regulatory gene of Escherichia coli polysaccharide synthesis gene (cps) and having a base sequence at cut position by a prescribed restriction enzyme (pstI, KpnI, etc.) in the rcsB gene region. The rcsB gene region is integrated into a plasmid pACYC184 to provide the objective plasmid.
