To provide a method for simplifying a peptide mixture by combining the protection of a first amino group and cation-exchange chromatography.
Selective separation of multiply charged peptides (RH peptides) of four on average per protein and the study of protein accounting for 90% of analyzed proteome are made possible. This method is suitable for quantitative proteomics study that does not use two-dimensional electrophoresis and is fit for any type of isotope label. When different isotope labels are used in a single experiment, this method is very useful for measuring the differential expression of proteins existing in a plurality of states (three to six states). In the chromatographic system used, separation/fractionation of RH peptide becomes possible, and this realizes identification of a greater number of proteins.
