PURPOSE: To provide a steroid derivative for preventing and treating diseases such as prostatic hypertrophy, prostatic cancer, etc., caused by excessive 5α- dihydrotestosterone, having excellent inhibitory action on testosterone 5α-reductase, comprising a steroid derivative.
CONSTITUTION: A corticosterone of formula I (Z is oxo, H and OH or H and 1-5C aliphatic acyloxy) is dissolved in dioxane, mixed with a hot aqueous solution of sodium periodate and stirred at 50°C for 1 day. The reaction solution is filtered, a water layer of the filtrate is acidified with hydrochloric acid, extracted with methylene chloride and treated with diazomethane to give an ester derivative of formula II (R5 is a carboxyl-protecting group). Then, the ester derivative is treated with dimethyl cyanophosphate, hydrolyzed, reacted with an amine of the formula HNR1R2 [R1 is H, 1-6C alkyl, etc.; R2 is a (substituted)alkyl, etc.] to give a compound of formula III. Further this compound is hydrolyzed or reduced to give the steroid derivative of formula IV [R3 is a (protected) COOH or (substituted)alkyl, etc.].
