Abstract:
PROBLEM TO BE SOLVED: To prepare a new compound useful as an antagonist of glycoproteins IIb/IIIa (GP IIb/IIIa) complex capable of inhibiting the blood platelet activation or aggregation and the treatment or prophylaxis of thrombosis or the like.
SOLUTION: This compound is represented by formula I (R1 is H, a 1-5C alkyl, a 3 8C cycloalkyl, an aralkyl, an alkoxycarbonyl or the like; A is methylene or ethylene; R is H, a 1-5C alkyl, an aryl or an aralkyl; Y is H, COOR2, an aryl, an aralkyl or the like; R2 is a 1-5C alkyl), e.g. methyl (4-{4-[4-(aminoiminomethyl)phenyl]-1,3-thiazol-2-ylamino}piperid-1-yl)acetate. The compound represented by formula I is prepared by reacting a compound represented by formula II in an alkanol in an acidic medium, then carrying out the Pinner reaction of the resultant compound, providing an imidate salt and subsequently reacting the obtained imidate salt with an amine such as ammonia (when Y is H). The compound is useful in the prophylaxis of peri- and post-operative complications consecutive to a heart and kidney, transplant and the treatment of osteoporosis, chronic rheumatoid arthritis and cancer.
