Abstract:
To identify a variety of sugar chains with improving the throughput of measurement.SOLUTION: After fluorescently labeling a sample of a sugar chain mixture (S2), fluorescence detection by separating it by a microchip electrophoresis method under a plurality of types of buffers with the addition of buffer and mutually different lectins without addition of lectins respectively (S3). A high concentration of gel with which a molecular sieve effect is obtained is used as the buffer. A plurality of electroferrograms are created based on the detection results (S4). The peak corresponding to the sugar chain is substantially lost since the movement of the sugar chain to which lectin is specifically bound is delayed. Therefore, the sugar chain is identified by estimating the structure of the sugar chain in the sample (S5) based on the presence or absence of peaks on the type of the lectin and a plurality of electroferrograms. The types of the sugar chains that can be identified also increases since the number of the types of available lectins greatly increases by using a high concentration of appropriate types of gel in the buffer. In addition, it is possible to improve the throughput because the measurement time is short compared to the capillary electrophoresis.SELECTED DRAWING: Figure 1
